Approximately what is the RPM of the engine at 45 MPH? So many variables, fuel supply, timing, temperature... Make sure you are using good gasoline. This is step 1.
Not enough octane. Not sure what your ACTUAL static compression is, but that cam may have enough dynamic compression to put you close to RACE gas territory.
I used that cam in a 215. A little too much there, but was just right in a Rover 4.0. I'd use the 50234 in the 340. We used the 50233 in a 300 stroker. It was pretty mild in that. You need lower Dynamic compression. There is 4 degree advance built in the cam. Try retarding it 4 degrees.
had the same thing going on with my 340 build after i recurved the distributor. think the dish pistons in these motors add to the problem. 30*at wot isn't very much timing for a 340. had to go with 10-12* initial coming all in at 2700-2800. with 32*-33* total if you can get there.
I would run a data log for timing, AFR, MAP & RPM when knocking occurs to see if your lean in that area and what actual timing is. Then you can correct what you feel is needed. Just don’t forget to save current config file before changing anything or you’ll learn why the hard way as I did. I’m by far not an expert. Good luck.
Are you running an automatic with overdrive and a lockup torque converter? If so, cruising at 45 mph you’re in OD and if you step into the torque converter needs to unlock or it’s gonna lug and detonate. Just a thought. First step is to turn on data logging and look at the numbers.
